The majora2–6 sialoglycoproteins in detergent-extracts of Kurloff cells were purified by anion-exchange andSambucus nigra agglutinin-affinity chromatographies. The similar ultrastructural localisations of (1)S. nigra agglutinin-gold conjugates and (2) acid phosphatase activities on the Kurloff body and particularly on its myelin figures indicated that the majora2-6 sialoglycoproteins of the Kurloff cell had acid phosphatase activity. Two-dimensional electrophoresis showed that these tartrate-sensitive phosphatases corresponded to 2 acidic (pI 3.4–3.7) polypeptides of 36 and 34 kDa. Hydrolysis with peptide-N-glycosidases F gave a 33 kDa apoprotein rich in alanine, glutamic acid, tyrosine and lysin. A lectin-affinity study demonstrated that they contained hybrid type bisected and fucosylatedN-linked oligosaccharides. Cytotoxic properties were previously attributed to Kurloff cells and other studies suggested that not only acid phosphatases but alsoa2-6-linked sialic acid residues themselves may participate in natural killer activity.  相似文献

Abstract Multivariate methods for the detection of impacts on communities are: (i) explored in the context of a disturbance experiment at a site of previous uranium mining in Kakadu National Park, Northern Territory; and (ii) applied to the design of a monitoring programme for a proposed mining development at nearby Coronation Hill. The disturbance experiment in Rockhole Mine Creek used a gravity-fed redirection of polluted water from the mine adit to create a point-source pollutant. Weekly samples of the macroinvertebrate communities were taken both before and after disturbance at the control and‘impact’ site, as part of a multivariate BACIP (Before-After-Control-Impact-Pairs) design. The results of the experiment show that the multivariate approach provided high sensitivity in detecting impacts on the community, with little difference in sensitivity between species and family level analyses. A simple procedure for deleting those taxa that show poor discrimination between pristine and presently disturbed areas further increased the sensitivity of the tests. In addition to the BACIP analyses, robust ordination of the community data suggested a time/treatment interaction effect. Application of the multivariate BACIP approach also was explored using baseline data for benthic communities from the South Alligator River near Coronation Hill. The method provides high statistical power for monitoring; a relatively small change in community dissimilarity would be detectable in a programme with 5 years of observations before and after commencement of mining. A robust ordination of the yearly samples for the control and potential-impact sites showed that, although samples varied along identifiable gradients over time, the two areas changed in parallel so that the difference between the areas was approximately constant over years. This result demonstrates that the multivariate BACIP approach can provide relative constancy of pre-impact difference values, a key requirement of BACIP designs that is difficult to satisfy using abundance data for a single species.  相似文献

Following the previous ultrastructural demonstration of the presence of arylsulphatase (Asase) activities in Kurloff cells (KC) and of their quasi-exclusive localization in the Kurloff body (KB), this work investigates their biochemical and zymographic properties after extraction from purified KC suspensions. Using the discriminative inhibitory conditions of both the Baum or LeeVaupel and Conzelmann methods, nitrocatechol sulphate hydrolyzing enzymes of the KC were assumed to belong to the B class of the type II Asase alone. After electrophoretic separation under non-denaturing conditions in a 4–23% polyacrylamide gel, they were characterized by 55 kDa and 62 kDa zymographic bands. After isoelectric focusing, ‘classical’ cationic isoforms (pI 8.5) and two anionic isoforms (pI 4.4 and 4.6) were observed on zymograms. As expected for class B Asase, the different zymographic forms of KC Asase were only recovered in the unadsorbed fraction after anion-exchange chromatography on DEAE-cellulose column equilibrated with high ionic strength buffer. Their Km (2.1 mM), their optimum pH (5.8) and their inhibitions by sulfite, phosphate, sulphate and ascorbic acid as well as their slight stimulation by AgNO3 were also characteristic of this class of Asase. Finally, chondroitin4-sulphate was shown to potentially be a physiological substrate for these lysosomal enzymes.  相似文献

Summary Classical theory states that cover of annual plants should increase relative to perennials as disturbance frequency increases. However, it has been suggested that long-distance clonal spreading can allow some perennial plants to survive in highly disturbed areas by quickly spreading into disturbed patches. To evaluate these hypotheses, we analysed data of plant distributions in two different ecosystems, a barrier island and a short-grass steppe. The disturbances studied were sand deposition during storms (overwash) on the barrier island and grazing by cattle in the short-grass steppe. In each case the disturbance frequency varied over the ecosystem; we categorized different areas in terms of their disturbance frequencies. All plant species in each area were categorized as one of four plant life forms (1) annual or biennial, (2) herbaceous perennial without long-distance clonal spreading (3) herbaceous perennial with long-distance clonal spreading (i.e guerilla form) and (4) woody plant. Percentage cover of each plant life form in each disturbance frequency category was calculated. In both ecosystems, (1) there was an increase in the relative cover of annuals as one moved from areas of low to moderate disturbance frequencies, but then a decrease in cover of annuals as one moved into the areas of highest disturbance frequency and (2) the guerilla forms showed the greatest relative increase in cover from moderately to highly disturbed areas. The combination of two factors can explain this pattern: (1) long-distance clonal spreading effectively reduces the time to colonization of recently disturbed sites and (2) effects of the disturbances in these two systems are probably more severe for seeds than for stems. We illustrate these effects using a spatially explicit simulation model of the population dynamics of plants in a disturbed landscape.  相似文献

Relationships between soil chemistry and population chemotype structure of Thymus pulegioides have been studied. The analysis of correlations suggest that an increased carbonate content in soil decreases the chemotype diversity of a population (as calculated by use of the Shannon index): the proportion of linalool chemotype plants rises and that of the phenol chemotype plants declines. In addition, the chemotype diversity decreases with increasing frequency of linalool chemotype, and increases with increase of carvacrol chemotype.  相似文献

It has been assumed that herbivores constitute a selective agent for the evolution of plant resistance. However, few studies have tested this hypothesis. In this study, we look at the annual weed Datura stramonium for evidence of current natural selection for resistance to herbivorous insects. Paternal half-sib families obtained through controlled crosses were exposed to herbivores under natural conditions. The plants were damaged by two folivorous insects: the tobacco flea beetle Epitrix parvula and the grasshopper Sphenarium purpurascens. Selection was estimated using a multiple-regression analysis of plant size and of damage by the two herbivores on plant fitness measured as fruit production for both individual phenotypes and family breeding values (genetic analysis). Directional phenotypic selection was detected for both larger plant size and lower resistance to the flea beetles, whereas stabilizing phenotypic selection was revealed for resistance to S. purpurascens. However, performing the same analyses on the breeding values of the characters revealed directional and stabilizing selection only for plant size. Thus, no agreement existed between the results of the two types of analyses, nor was there any detectable potential for genetic change in the studied population because of selection on herbivore resistance. The narrow-sense heritability of every trait studied was small (all <0.1) and not different from zero. The potential for evolutionary response to natural selection for higher resistance to herbivores in the studied population of D. stramonium is probably limited by lack of genetic variation. Natural selection acts on phenotypes, and the detection of phenotypic selection on resistance to herbivores confirms their ecological importance in determining plant fitness. However, evolutionary inferences based solely on phenotypic selection analyses must be interpreted with caution.  相似文献

Substitution processes are of two sorts: origination processes record the times at which nucleotide mutations that ultimately fix in the population first appear, and fixation processes record the times at which they actually fix. Substitution processes may be generated by combining models of population genetics—here the symmetrical-neutral, overdominance, underdominance, TIM, and SAS-CFF models—with the infinite-sites, no-recombination model of the gene. This paper is mainly concerned with a computer simulation study of these substitution processes. The rate of substitution is shown to be remarkably insensitive to the strength of selection for models with strong balancing selection caused by the genealogical drift of mutations through alleles held in the population by selection. The origination process is shown to be more regular than Poisson for the overdominance, TIM, and SAS-CFF models but more clustered for the underdominance model. A class of point processes called Sawyer processes is introduced to help explain these observations as well as the observation that the times between successive originations are nearly uncorrelated. Fixation processes are shown to be more complex than origination processes, with regularly spaced bursts of multiple fixations. An approximation to the fixation process is described. One important conclusion is that protein evolution is not easily reconciled with any of these models without adding perturbations that recur on a time scale that is commensurate with that of molecular evolution.  相似文献

Canalization is an abstract term that describes unknown developmental mechanisms that reduce phenotypic variation. A trait can be canalized against environmental perturbations (e.g., changes in temperature or nutrient quality), or genetic perturbations (e.g., mutations or recombination); this paper is about genetic canalization. Stabilizing selection should improve the canalization of traits, and the degree of canalization should be positively correlated with the traits' impact on fitness. Experiments testing this idea should measure the canalization of a series of traits whose impact on fitness is known or can be inferred, exclude differences among traits in the number of loci and alleles segregating as an explanation for the pattern of variability found, and distinguish between canalization against genetic and environmental variation. These conditions were met by three experiments within which the variation of fitness components among Drosophila melanogaster lines was measured and among which the genetic contribution to the variation among lines was clearly different. The canalization of the traits increased with their impact on fitness and did not depend on the degree of genetic differences among lines. That the flies used had been transformed by a P-element insert suggests that canalization was also effective against novel genetic variation. The results reported here cannot be explained by the classical hypothesis of reduction in the number of loci segregating for traits with greater impact on fitness and confirm that traits with greater impact on fitness are more strongly canalized. This pattern of canalization reveals an underappreciated role for development in microevolution. There is differential genetic canalization of fitness components in D. melanogaster.  相似文献
